When you add, move, or remove a stop, the route drawn on your map is briefly out of date until you rebuild it. That is all this prompt means.
Before a trip has ever been routed, the button reads Calculate Best Route. After an edit that changes the shape of the trip, it reads Recalculate Route. Editing that only changes notes does not make the route stale, so you will not be nudged for those.
Recomputing redraws the route and updates the distances and driving times. It does not throw away stops you have already resolved, so a campground you picked stays picked.
